By sequencing clones from a size-fractionated fetal brain cDNA library, Nagase et al. (2001) cloned ABLIM2, which they designated KIAA1808. The deduced amino acid sequence shares significant similarity with ABLIM1 (602330). RT-PCR ELISA detected high expression in adult brain and skeletal muscle, low expression in adult testis and ovary and fetal liver, and moderate expression in all other adult tissues and specific adult brain regions examined and in fetal brain. Klimov et al. (2005) identified 2 variants of ABLIM2 that differed in the presence or absence of coding exons 16 and 17. The deduced full-length 611-amino acid protein has 4 N-terminal LIM domains and a C-terminal villin (VIL; 193040) headpiece (VHP) domain predicted to mediate actin binding. RT-PCR detected ABLIM2 expression in blood, breast, kidney, cervix, and ovary. Klimov et al. (2005) also identified 2 Ablim2 variants in mouse and rat. In rat, highest Ablim2 mRNA was detected in brain and eye, with trace amounts in other tissues examined. Both Ablim2 variants were detected in mouse embryo. Using mouse Stars (ABRA; 609747) as bait in a yeast 2-hybrid screen of a human skeletal muscle CDNA library, followed by EST database analysis and PCR of skeletal muscle, cardiac, and brain cDNA, Barrientos et al. (2007) cloned 2 variants of ABLIM2. The deduced proteins contain 649 and 559 amino acids. Northern blot analysis detected highest expression in skeletal muscle and lower expression in brain, spleen, and kidney. Heart and skeletal muscle expressed both ABLIM2 variants. In situ hybridization of embryonic day-15.5 mouse showed high Ablim2 expression in skeletal muscle and weaker expression in the central nervous system. Ablim2 and Ablim3 (611305) showed different expression patterns in regions of adult mouse brain, including hippocampus and cerebellum. Immunohistochemical analysis of mouse hindlimb revealed a striated pattern of Ablim2 staining, consistent with a sarcomeric or costameric localization.
