By PCR of human macrophage cDNA using degenerate primers homologous to human ABCA6 (612504) and ABCA8 (612505), Piehler et al. (2002) cloned ABCA9. The deduced 1,624-amino acid protein has a calculated molecular mass of about 160 kD. It has 2 nucleotide-binding folds, each with characteristic Walker A and B motifs, and 2 transmembrane domains, each consisting of 7 membrane-spanning segments. ABCA9 shares 60% and 72% amino acid identity with ABCA6 and ABCA8, respectively, and like those proteins, it contains an ABC transporter signature sequence only in its N-terminal nucleotide-binding fold. Piehler et al. (2002) also identified 3 splice variants of ABCA9 that encode proteins of 266, 1,435, and 1,440 amino acids. The shortest protein includes only 2 N-terminal transmembrane segments, and the other proteins are truncated within the C-terminal nucleotide-binding fold. RNA dot blot analysis detected ABCA9 expression in all adult and fetal tissues examined, with highest levels in adult heart, brain, and liver.
