NOS3 (Nitric Oxide Synthase 3) is a protein coding gene located on human chromosome 7q36.1. NOS3 is also known as eNOS and ECNOS. The human NOS3 gene encodes a 133275 Da protein containing 1203 amino acids. The NOS3 protein is biasedly expressed in spleen, placenta and other tissues. Among its related pathways are HIF1Alpha Pathway and Signaling by GPCR. NOS3 is related to oxidoreductase activity and iron ion binding. NOS1 is an important paralog of NOS3 gene. NOS3 is associated with some diseases, including Alzheimer Disease and Stroke, Ischemic.

    Nos3, a gene regulated by many congenital heart disease risk factors including stress and diabetes, interacts genetically with Tbx5 provides a molecular framework to understand gene-environment interaction in the setting of human birth defects. The Nos3 gene is known to regulate vascular tone via the endothelial nitric oxide synthase/nitric oxide pathway. Nos3as NAT is known to act as a negative regulator of Nos3 gene expression.
