NKG2D/A: Emerging Immunotherapy Checkpoint Targets

NKG2D and NKG2A belong to the NKG2 (CD159) family. NKG2 proteins are predominantly expressed on cytotoxic immune cells. They are new attractive targets for the development of immune checkpoint modulators.

NKG2A is an inhibitory receptor. It forms a heterodimer with CD94 (KLRD1). CD94/NKG2A complex interacts with the non-classical MHC class I molecule HLA-E on target cells. This interaction inhibits the activation of NK cells and T cells via the tyrosine phosphatases SHP-1 or SHP-2 signaling cascades. Similar to other immune checkpoints (such as PD-1/PD-L1), CD94/NKG2A-HLA-E is also used by cancer cells to escape the immune system.

NKG2D is an activating receptor. The activation of NKG2D by its ligands, including MICA, MICB, and ULBPs (ULBP1, ULBP2, ULBP3, ULBP4, ULBP5, ULBP6) in human and H60 (a–c), RAE(α–ε) and MULT1 families in mice, is an important anti-tumor signaling axis. In mice, NKG2D uses adaptor molecules DAP10 and DAP12 to relay its signal transduction, while in human, NKG2D associates exclusively with DAP10. It can also regulate cytokine production (e.g. IL-10, IL-9, IL-13). NKG2D also plays an important role against virus infection.
