Immunofluorescence staining of PTBP1 in HeLa cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-human PTBP1 polyclonal antibody (1:1000) at 4℃ overnight. Then cells were stained with the Alexa Fluor®594-conjugated Goat Anti-rabbit IgG secondary antibody (red)Positive staining was localized to nucleus.

PTBP1 Background Information

Polypyrimidine tract-binding protein 1 (PTBP1) is a highly conserved RNA-binding protein that is a well-known regulator of alternative splicing. PTBP1, a heterogeneous nuclear ribonucleoprotein, is a multifunctional RNA-binding protein. PTBP1 participates in some biological processes, including maintaining cell structure and motility, immunity, protein metabolism, and the cell cycle. PTBP1 involving in almost all steps of mRNA regulation including alternative splicing metabolism during tumorigenesis due to its RNA-binding activity. PTBP1 facilitates colorectal cancer migration and invasion activities by the inclusion of cortactin exon 11 and contributes to spermatogenesis through the regulation of spermatogonia proliferation. It has been reported that PTBP1 is overexpressed in many different types of cancer and this high expression is associated with increased proliferation and poor prognoses.
