Immunofluorescence staining of TUBA4A in HeLa cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti- TUBA4A polyclonal antibody (1:1000) at 4℃ overnight. Then cells were stained with the Alexa Fluor®594-conjugated Goat Anti-rabbit IgG secondary antibody (red) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

TUBA4A was immunoprecipitated using:

Lane A:0.5 mg K562 Whole Cell Lysate

4 µL anti-TUBA4A rabbit polyclonal antibody and 15 μl of 50 % Protein G agarose.

TUBA4A Background Information

TUBA4A gene has recently been identified as a potential candidate amyotrophic lateral sclerosis(ALS)-associated gene using exome-wide rare variant burden analysis. TUBA4A mutants destabilize the microtubule network, diminishing its repolymerization capability. Which further emphasize the role of cytoskeletal defects in ALS and demonstrate the power of gene-based rare variant analyses in situations where causal genes cannot be identified through traditional segregation analysis.
