In the synthesis of class II antigens of the major histocompatibility complex, newly synthesized alpha and beta chains form complexes with gamma chains in the endoplasmic reticulum. After transport of the complex to the Golgi apparatus, and concomitant with terminal glycosylation, the gamma chain dissociates from the other 2 chains. Subsequently, at least some of the gamma chains become integrated into the plasma membrane in a transmembrane position. Claesson et al. (1983) isolated a cDNA encoding the human gamma chain from a Raji lymphoblastoid cell cDNA library. The deduced 216-amino acid protein contains a short N-terminal cytoplasmic segment, followed by a transmembrane region and an extracellular C-terminal segment. It lacks an N-terminal signal sequence. In vitro-translated human gamma chain had an apparent molecular mass of 33 kD by SDS-PAGE. Schori et al. (2007) stated that mouse Cd74 is a nonpolymorphic type II integral membrane protein with an N-terminal 28-amino acid cytoplasmic tail, a 24-amino acid transmembrane region, and a 150-amino acid luminal domain. Differential splicing results in 31- and 41-kD isoforms designated p31 and p41, respectively.
