0.5 mg/mL of mouse anti-Human TNF-alpha / TNFA / TNFSF2 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2 μg/mL in PBS before coating.
0.2 mg/mL of mouse anti-Human TNF-alpha / TNFA / TNFSF2 monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4, store at 4℃). Dilute to working concentration of 1.5 μg/mL in detection antibody dilution buffer before use.
Each vial contains 45 ng of recombinant Human TNF-alpha / TNFA / TNFSF2. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 600 pg/mL is recommended.
The minimum detectable dose of Human TNF-alpha / TNFA / TNFSF2 was determined to be approximately 9.38 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.
|The Human TNF-alpha / TNFA / TNFSF2 ELISA Pair Set is for the quantitative determination of Human TNF-alpha / TNFA / TNFSF2.
The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for Human TNF-alpha / TNFA / TNFSF2 coated on a 96-well plate. Standards and samples are added to the wells, and any Human TNF-alpha / TNFA / TNFSF2 present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated mouse anti-Human TNF-alpha / TNFA / TNFSF2 monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of Human TNF-alpha / TNFA / TNFSF2 present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

Tumor necrosis factor alpha (TNF-alpha), also known as TNF, TNFA or TNFSF2, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. Two receptors, TNF-R1 (TNF receptor type 1; CD120a; p55/60) and TNF-R2 (TNF receptor type 2; CD120b; p75/80), bind to TNF-alpha. TNF-alpha protein is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-alpha is involved in fighting against the tumorigenesis, thus, is regarded as a molecular insight in cancer treatment.
