Human SCGN Materials provided
Capture Ab:0.1 mg/mL of mouse anti-SCGN monoclonal antibody. Dilute to a working concentration of 2.0 μg/mL in CBS before coating.
Detection Ab:0.5 mg/mL mouse anti-SCGN monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 1 μg/mL in detection antibody dilution buffer before use.
Standard:Each vial contains 21 ng of recombinant SCGN. Reconstitute standard powder with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 500 pg/mL is recommended.
Human SCGN Specificity
Human SCGN Sensitivity
The minimum detectable dose of Human SCGN / Secretagogin was determined to be approximately 7.8 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.
Human SCGN Principle of the product
The Human SCGN / Secretagogin ELISA Pair Set is for the quantitative determination of Human SCGN / Secretagogin.
This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for SCGN / Secretagogin coated on a 96-well plate. Standards and samples are added to the wells, and any SCGN / Secretagogin present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated mouse anti-SCGN / Secretagogin monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of SCGN / Secretagogin present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.
Human SCGN Storage

SCGN / Secretagogin Background

Secretagogin, also known as SCGN, is a secreted protein which is detectable in human serum after ischemic neuronal damage. It is a recently described calcium-binding protein. Secretagogin / SCGN is expressed at high levels in the pancreatic islets of Langerhans and to a much lesser extent in the gastrointestinal tract (stomach, small intestine and colon), the adrenal medulla and cortex and the thyroid C-cells. In the brain, the expression of Secretagogin / SCGN is restricted to distinct subtypes of neurons with highest expression in the molecular layer of the cerebellum (stellate and basket cells), in the anterior part of the pituitary gland, in the thalamus, in the hypothalamus and in a subgroup of neocortical neurons. Secretagogin / SCGN is widely expressed in prostatic adenocarcinoma as opposed to adenocarcinomas in other organs. The function of Secretagogin / SCGN is unknown, but it has been suggested in beta-cells to influence calcium-influx and has been observed downregulated in diabetes-prone BB rat islets exposed to cytokines. Secretagogin / SCGN is involved in the calcium metabolism of tumour cells and endothelial cells in a subset of neoplasms of the brain and its coverings. Secretagogin / SCGN is also a novel marker for neuroendocrine differentiation.
