Standards and samples are added to the wells,and any TNFRI / CD120a present binds to the immobilized antibody. The wells arewashed and a horseradish peroxidase conjugated mouse anti-TNFRI / CD120amonoclonal antibody is then added, producing an antibody-antigen-antibody"sandwich". The wells are again washed and TMB substrate solution is loaded,which produces color in proportion to the amount of TNFRI / CD120a present inthe sample. To end the enzyme reaction, the stop solution is added andabsorbances of the microwell are read at 450 nm.

CD120a (cluste of differentiation 120a), also known as TNFR1 / TNFRSF1A, is a member of CD family, tumor necrosis factor receptor superfamily. CD120a is one of the most primary receptors for the tumor necrosis factor-alpha. It has been shown to be localized to both plasma membrane lipid rafts and the trans golgi complex with the help of the death domain (DD). CD120a can activate the transcription factor NF-κB, mediate apoptosis, and regulate inflammation processes.
