Human ECM1 Materials provided
Capture Ab:0.5 mg/mL of mouse anti-ECM1 monoclonal antibody. Dilute to a working concentration of 2.0 μg/mL in CBS before coating.
Detection Ab:0.6 mg/mL rabbit anti-ECM1 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.5 μg/mL in detection antibody dilution buffer before use
Standard:Each vial contains 110 ng of recombinant ECM1. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 1500 pg/mL is recommended.

Human ECM1 Principle of the product
The human ECM1 ELISA Pair Set is for the quantitative determination of human ECM1.
This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for ECM1coated on a 96-well plate. Standards and samples are added to the wells, andany ECM1 present binds to the immobilized antibody. The wells are washed and ahorseradish peroxidase conjugated rabbit anti-ECM1 monoclonal antibody is thenadded, producing an antibody-antigen-antibody "sandwich". The wells are againwashed and TMB substrate solution is loaded, which produces color in proportionto the amount of ECM1 present in the sample. To end the enzyme reaction, thestop solution is added and absorbances of the microwell are read at 450 nm.

Capture Antibody: Aliquot and store at -20℃ to -80℃ for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Protect it from prolonged exposure to light. Aliquot and store at -20℃ to -80℃ and for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Standard: Store lyophilized Standard at -20℃ to -80℃ for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80℃ for up to 1 month. Avoid repeated freeze-thaw cycles.

ECM1  Background

Extracellular matrix protein 1 (ECM1) is a secreted glycoprotein and playing a pivotal role in endochondral bone formation, angiogenesis, and tumour biology. Three splice variants have been identified: ECM1a (540 aa) is most widely expressed, with highest expression in the placenta and heart; ECM1b (415 aa) is differentiation-dependent expressed and found only in tonsil and associated with suprabasal keratinocytes; ECM1c (559 aa) accounts for approximately 15% of skin ECM1. Although ECM1 is not tumor specific, is significantly elevated in many malignant epithelial tumors and is suggested as a possible trigger for angiogenesis, tumor progression and malignancies. It also has been shown to regulate endochondral bone formation, skeletal development and tissue remodeling.
