Human CDH1 Materials provided
Capture Ab:0.4 mg/mL of mouse anti-Cadherin-1 monoclonal antibody. Dilute to a working concentration of 2 μg/mL in CBS before coating.
Detection Ab:0.5 mg/mL of biotinylated rabbit anti-Cadherin-1 polyclonal antibody. Dilute to a working concentration of 0.3 μg/mL in detection antibody dilution buffer before use.
Standard:Each vial contains 100 ng of recombinant Cadherin-1. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 3 ng/mL is recommended.
Streptavidin-HRP:50 μL of streptavidin conjugated to horseradish-peroxidase. 1:2000 Dilution in detection antibody dilution buffer before use.

The minimum detectable dose of human Cadherin-1 (E-cadherin / CDH1 / CD324) was determined to be approximately 46.8 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

The human Cadherin-1 ELISA Pair Set is for the quantitative determination of human Cadherin-1.
This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for Cadherin-1(E-cadherin / CDH1 /CD324) coated on a 96-wellplate. Standards and samples are added to the wells, and any Cadherin-1 presentbinds to the immobilized antibody. The wells are washed and a biotinylatedrabbit anti-Cadherin-1 polyclonal antibody is then added,producing an antibody-antigen-antibody "sandwich". To produces color inproportion to the amount of Cadherin-1 present in the sample strepavidin-HRPand TMB substrate solution are loaded. The absorbances of the microwell areread at 450 nm.

Capture Antibody: Aliquot and store at -20℃ to -80℃ for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Aliquot and store at -20℃ to -80℃ for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Standard: Store lyophilized Standard at -20℃ to -80℃ for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80℃ for up to 1 month. Avoid repeated freeze-thaw cycles.
Streptavidin-HRP: Store at 4℃ and protect it from prolonged exposure to light. DO NOT FREEZE! It is stable for up to 6 months from date of receipt.

E-Cadherin/CDH1/E-cad/CD324 Background

Cadherins are calcium-dependent cell adhesion proteins which preferentially interact with themselves in a homophilic manner in connecting cells, and thus may contribute to the sorting of heterogeneous cell type. E-cadherin (E-Cad), also known as CDH1 and CD324, is a calcium-dependent cell adhesion molecule the intact function of which is crucial for the establishment and maintenance of epithelial tissue polarity and structural integrity. Mutations in CDH1 occur in diffuse type gastric cancer, lobular breast cancer, and endometrial cancer. In human cancers, partial or complete loss of E-cadherin expression correlates with malignancy. During apoptosis or with calcium influx, E-Cad is cleaved by the metalloproteinase to produce fragments of about 38 kDa (E-CAD/CTF1), 33 kDa (E-CAD/CTF2) and 29 kDa (E-CAD/CTF3), respectively. E-Cad has been identified as a potent invasive suppressor, as downregulation of E-cadherin expression is involved in dysfunction of the cell-cell adhesion system, and often correlates with strong invasive potential and poor prognosis of human carcinomas.
