VEGFR3 / FLT-4 Antibody Background
VEGF R3 (Flt-4), together with the other two members VEGFR1 (Flt-1) and VEGFR2 (KDR/Flk-1) are receptors for vascular endothelial growth factors (VEGF) and belong to the class III subfamily of receptor tyrosine kinases (RTKs). They share the similar structure characteristics of seven immunoglobulin-like repeats in their extracellular domain, a single transmembrane spanning region and an intracellular portion containing a split tyrosine-kinase domain. The fifth Ig domain of VEGFR3 ECD is proteolytically cleaved and the resulting polypeptides remain linked by two disulfide-bonds. VEGFR3/FLT-4 is widely expressed in the early embryo but becomes restricted to lymphatic endothelia at later stages of development. Upon ligand binding, VEGFR3/FLT-4 become activated through transphosphorylation after homodimerization and subsequently interacts with the adaptor proteins such as SHC and GRB2, and thus mediates the cellular responses of VEGF. Both VEGF-C and VEGF-D have been identified as ligands for VEGF R3. The VEGF receptors are essential for vasculogenesis and angiogenesis, while VEGFR3/FLT-4 plays a significant role in ligand-mediated lymphangiogenesis. Mutation in this gene is associated with the primary lymphedema which is a rare genetic condition with both autosomal dominant and autosomal recessive modes of inheritance.
