Tie2 / TEK protein

Bio-activity:

Measured by its binding ability in a functional ELISA.
Immobilized mouse TEK-His (Cat: 51087-M08H) at 10 μg/ml (100 μl/well) can bind human Ang2-Fc (Cat: 10691-H02H) with a linear range of 6.25-200 ng/ml.
Endotoxin: < 1.0 EU per μg of the protein as determined by the LAL method
Stability: Samples are stable for up to twelve months from date of receipt at -70℃
Predicted N terminal: Val 19
Molecular Mass:

The recombinant mouse TEK comprises 737 amino acids and has a predicted molecular mass of 82.4 kDa. The apparent molecular mass of the protein is approximately 91 kDa in SDS-PAGE under reducing conditions due to glycosylation.

Formulation: Lyophilized from sterile PBS, pH7.4.
  1. Normally 5 % - 8 % trehalose and mannitol are added as protectants before lyophilization. Specific concentrations are included in the hardcopy of COA.
  2. Please contact us for any concerns or special requirements.

Tie2 / TEK Protein Usage Guide

Storage: Store it under sterile conditions at -70℃. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
Reconstitution: A hardcopy of COA with reconstitution instruction is sent along with the products. Please refer to it for detailed information.

Tie2 / TEK Protein Description

Tie2/Tek, together with Tie1/Tie, are members of the receptor tyrosine kinase (RTK) subfamily, and possess a unique extracellular domain containing two immunoglobulin-like loop domains flanking three epidermal growth factor (EGF) repeats followed by three fibronectin-type I II repeats, and a split tyrosine kinase domain in the cytoplasmic region. Tie2 is expressed predominantly on endothelial and hematopoietic progenitor cells and is essential for blood vessel formation and maintenance. The identified ligand angiopoietin-1 (Ang-1) binds Tie2 receptor with high-affinity and induces the signaling pathway of both cell migration and cell survival via the related mediator such as GRB2 and SH-PTP2. Accordingly, the Tie2 may trigger crucial endothelial responses during embryonic vascular development and during pathologic vascular growth, as well as the inflammatory responses with the key regulator ABIN-2. Recent study has revealed that Ang1-Tie2 signaling exerts different function at cell-cell contacts and cell-substratum contacts dependent upon the spatial localization of Tie2. Deficiency in this gene is associated with inherited venous malformations.
