Background
TNF-related apoptosis-inducing ligand (TRAIL), also known as Apo-2 ligand and TNFSF10, are TNF superfamily members, and preferentially induces apoptosis in tumor cell lines, but not in normal cells. TRAIL binds at least five different receptors with characteristic cysteine-rich repeats. Human TRAILR4, also known as decoy receptor 2 (DcR2) and TRUNND (TRAIL receptor with a truncated death domain), is a member of the TNF receptor superfamily and is referred to as TNFRSF10D. TRAILR4 shows a unique expression pattern in human tissues and is particularly abundant in fetal liver and adult testis. This protein is a type 1 transmembrane receptor consisting of an extracellular TRAIL-binding domain, a transmembrane domain, and a truncated cytoplamic death domain. Accordingly, TRAILR4 is not capable of activating TRAIL-mediated NF-kappaB or apoptosis pathways, and identified as an antiapoptotic receptor, although it retains one third of a consensus death domain motif in its cytoplasmic domain. It has been demonstrated that TRAILR4 expression is directly regulated by p53 and attenuates cell response to DNA-damaging agents.
