> TLE3 TLE3
TLE3 (transducin-like enhancer of split 3), and several other members of the Gro/TLE family of groucho-related genes are expressed during pituitary development. The transducin-like enhancers of split are a family of mammalian proteins that share sequence homology with the Drosophila protein Groucho. TLE1 and TLE3 a have similar protein structure, and their patterns of expression during pituitary development are overlapping, but distinct. The groucho family member TLE3 is a transcriptional integrator of the PPAR? and Wnt pathways. TLE3 might also play roles independent of HESX1 by interacting with other transcription factors like PROP1.

Summaries for TLE3
Wikipedia summary for TLE3:
Transducin-like enhancer protein 3 is a protein that in humans is encoded by the TLE3 gene.
Human TLE3 Protein General Information
| Protein names |
Transducin-like enhancer protein 3 |
| Sequence length |
772 AA. |
| Domain |
Repeat
|
| Sequence similarities: |
Belongs to the WD repeat Groucho/TLE family. |
| Post-translational modification: |
Ubiquitinated by XIAP/BIRC4. This ubiquitination does not affect its stability, nuclear localization, or capacity to tetramerize but inhibits its interaction with TCF7L2/TCF4. |
| Subunit structure |
Homotetramer and heterooligomer with other family members. Binds LEF1, TCF7 and TCF7L1 By similarity. Binds FOXA2. Interacts with XIAP/BIRC4 and TCF7L2/TCF4. |
| Subcellular location: | Nucleus. |
| Tissue specificity |
Placenta and lung. |
| Sequence caution | The sequence BAB13373.1 differs from that shown. Reason: Erroneous initiation. |

Function for TLE3 Protein
UniProtKB:
TLE3 is transcriptional corepressor that binds to a number of transcription factors. TLE3 inhibits the transcriptional activation mediated by CTNNB1 and TCF family members in Wnt signaling. The effects of full-length TLE family members may be modulated by association with dominant-negative AES.
Genatlas:
- TLE3 is transcriptional corepressor binding to a number of transcription factors
- TLE3 inhibits the transcriptional activation mediated by CTNNB1 and TCF family members in Wnt signaling
- TLE3 might also play roles independent of HESX1 by interacting with other transcription factors like PROP1 (Carvalho 2010)
Homology for human TLE3
- homolog to Drosophila split Groucho (Notch signaling pathway)
- homolog to murine Tle3 (98.8pc)
- homolog to rattus Tle3 (99.2pc)
