TIMP-1 / TIMP1 Antibody Usage Guide

Specificity :

Human TIMP1

  No cross-reactivity in ELISA with

Human TIMP2

Western blot : This antibody can be used at 1 - 2 μg/mL with the appropriate secondary reagents to detect human TIMP1 in WB. Using a DAB  detection system, the detection limit for human TIMP1 is approximately 1 ng/lane under non-reducing conditions and 10 ng/lane under reducing conditions
Direct ELISA : This antibody can be used at 0.1 - 0.2 μg/mL with the appropriate secondary reagents to detect human TIMP1. The detection limit for human TIMP1 is approximately  0.00975 ng/well
Storage : This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

TIMP-1 / TIMP1 Antibody Background

Tissue inhibitors of metalloproteinases (TIMP) family are natural inhibitors of the matrix metalloproteinases (MMPs), the zinc enzymes involved in extracellular matrix maintenance and remodeling. The TIMP family encompasses four members (TIMP1-4), and the inducible form TIMP-1/TIMP1 is a 28 kDa glycoprotein expressed by a variety of cell types. It inhibits most MMPs except membrane-type MMP subfamily by forming non-covalent binary complex, but also interacts with the hemopexin-like (PEX) domain of pro MMP-9. TIMP-1/TIMP1 has been identified as a multifunctional molecule with divergent functions. It exhibits keratinocyte and fibroblast growth factor-like activity and acts as a cell survival factor, and additionally participates in tumor metastasis, angiogenesis, inflammatory responses, as well as CNS homeostasis. Thus, TIMP-1/TIMP1 overexpression may serve to help identify patients with particularly aggressive disease for adjuvant treatments.
