SerpinA1 / A1AT Protein Description
SerpinA1, also known as Alpha-1 antitrypsin (A1AT), is a prototype member of the Serpin superfamily of the serine protease inhibitors. SerpinA1 is synthesized in hepatocytes, and to a lesser extent, in macrophages as well as intestinal epithelial cell lines. SerpinA1 is secreted as the abundant proteinase inhibitor in the circulation whose targets include elastase, plasmin, thrombin, trypsin, chymotrypsin, and plasminogen activator. Point mutations in the native SerpinA1 variants result in SerpinA1 deficiency, and consequently lead to severe lesions. For example, the well recognized S and Z alleles form polymers due to the conformational transition and are retained within the endoplasmic reticulum of hepatocytes. Accordingly, SerpinA1 deficiency in circulation is associated with emphysema or liver disease.
