SIRT1 Antibody Product Information
Recombinant Human SIRT1 protein (Catalog#11748-H07E)
|Antibody Type :||Mouse Monoclonal Antibody ( Mouse mAb Service Platform )|
Clone ID :
|Ig Type :||
|Formulation :||0.2 μm filtered solution in PBS with 5% trehalose|
This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human SIRT1 (rh SIRT1; Catalog#11748-H07E; NP_036370.2; Met 193-Ser 747). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.
SIRT1 Antibody Usage Guide
No cross-reactivity in ELISA with
E.coli cell lysate
|Western blot :||This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Human SIRT1 in WB. Using a DAB detection system, the detection limit for Human SIRT1 is approximately 8 ng/lane under reducing conditions. Use of this antibody under non-reducing conditions is not recommended.|
|Storage :||This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

SIRT1 Antibody Background
NAD-dependent deacetylase sirtuin-1, also known as SIR2-like protein 1, SIRT1 and SIR2L1, is a nucleus protein which belongs to thesirtuin family. SIRT1 / SIR2L1 is widely expressed and contains onedeacetylase sirtuin-type domain. Mammalian SIRT1 is an NAD-dependent deacetylase with critical roles in the maintenance of homeostasis and cell survival. Elevated levels of SIRT1 protein are evident in cancer in which SIRT1 can function as a cancer-specific survival factor. SIRT1 / SIR2L1 regulates processes such as apoptosis and muscle differentiation by deacetylating key proteins. It deacetylates 'Lys-382' of p53/TP53 and impairs its ability to induce proapoptotic program and modulate cell senescence. SIRT1 / SIR2L1 deacetylates TAF1B and thereby represses rDNA transcription by the RNA polymerase I. It is involved in HES1- and HEY2-mediated transcriptional repression. It inhibits skeletal muscle differentiation by deacetylating PCAF and MYOD1. SIRT1 / SIR2L1 may serve as a sensor of the cytosolic ratio of NAD+/NADH, which is essential in skeletal muscle cell differentiation. In case of HIV-1 infection, it interacts with and deacetylates the viral Tat protein.
