Preparation :

This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, human cell-derived, recombinant human RBP4 (rhRBP4; Catalog#10354-H08H; aa 1 - 201; NP_006735.2). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

RBP4 Antibody Usage Guide

Specificity :

Human RBP4 / PRBP

 

No cross-reactivity with human cell lysate ( 293 cell line ) in WB and ELISA.

Western blot : This antibody can be used at 1-2μg/mL with the appropriate secondary reagents to detect RBP4 in WB. Using a DAB detection system, the detection limit for human RBP4 is approximately 2ng/lane under non-reducing conditions and 100ng/lane under reduced conditions.
Direct ELISA : This antibody can be used at 0.5 - 1μg/mL with the appropriate secondary reagents to detect RBP4. The detection limit for RBP4 is approximately 1.25ng/well.
Storage : This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

RBP4 Antibody Background

Retinol-binding protein 4 (RBP4) is the specific carrier for retinol (also known as vitamin A), and is responsible for the conversion of unstable and insoluble retinol in aqueous solution into stable and soluble complex in plasma through their tight interaction. As a member of the lipocalin superfamily, RBP4 containing a β-barrel structure with a well-defined cavity is secreted from the liver, and in turn delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP4-retinol complex interacts with transthyretin (TTR), and this binding is crucial for preventing RBP4 excretion through the kidney glomeruli. RBP4 expressed from an ectopic source efficiently delivers retinol to the eyes, and its deficiency affects night vision largely. In addition, RBP4 is recently identified as an adipokine whose dysregulation is closely associated with the pathogenesis of insulin resistance and type 2 diabetes, as well as obesity.
