PTPRA / PTPalpha (aa 174-793) Protein Description

Protein tyrosine phosphatase, receptor type, A (PTPRA) also named PTPalpha, is a transmembrane member of the extensive family of protein-tyrosine phosphatases (PTPs). PTPRA/PTPalpha is constitutively phosphorylated on Tyr789, a consensus binding site for the SH2 domain of the SH3-SH2-SH3 adaptor protein GRB2. Human PTPRA/PTPalpha consists of 802 amino acids. The extracellular domain of 150 residues includes a hydrophobic signal peptide and eight potential N-glycosylation sites. This is followed by a transmembrane region and two tandemly repeated conserved domains. PTPRA/PTPalpha mediates NB-3 that plays an important role in the apical dendrite development of layer V pyramidal neurons in the visual cortex. Cortical neurons from PTPRA/PTPalpha knock-out mice exhibited a lower level of NB-3 at the cell surface. Co-expression of PTPRA/PTPalpha increased the cell surface distribution of NB-3. Further analysis showed that PTPRA/PTPalpha facilitated Golgi exit of NB-3 and stabilized NB-3 protein at the cell surface by preventing its release from the plasma membrane. The extracellular region of PTPRA/PTPalpha but not its catalytic activity is necessary for its effect on NB-3 expression.
