PTP1B / PTPN1 Antibody Background
Protein tyrosine phosphatase 1B (PTP1B) is the prototypic member of the PTP family which catalyze the hydrolysis of the phosphate monoesters specifically on tyrosine residues, and is identified based on the enzymatic activity and amino acid sequence. PTP1B/PTPN1 is a ubiquitously expressed non-transmembrane enzyme which resides in the endoplasmic reticulum with its conserved catalytic regions exposed to the cytosol. PTP1B/PTPN1 is reported to dephosphorylates a variety of substrates including receptor tyrosine kinases (RTKs) on the cell surface transmitting critical intracellular signals, as well as JAK2, Tyk2 and STAT5a/b, and thus plays a key role in negatively regulating the multiple tyrosine phosphorylation-dependent signaling pathways. Accordingly, this enzyme is implicated in the regulation of cell growth, differentiation, cell responses to cytokines and hormones, and malignancy transformation. In addition, it has been demonstrated that the CLK kinases activate PTP1B/PTPN1 can be activated by the CLK kinases. PTP1B/PTPN1 is now suggested to be a novel therapeutic target for the treatment of type 2 diabetes, obesity, and other associated metabolic syndromes.
