PPM1A / PP2C-alpha Antibody Background
Mouse Protein phosphatase 1A, also known as Protein phosphatase 2C isoform alpha, PP2C-alpha and Protein phosphatase IA and PPM1A, is a member of the PP2C family. PPM1A / PP2C-alpha is a phosphatase which dephosphorylates and inactivates a broad range of substrates, including TGF-beta, MAP kinases, p38 and JNK kinase cascades, and is involved in tumor suppression. PPM1A dephosphorylates and promotes nuclear export of TGFbeta-activated Smad2/3. Ectopic expression of PPM1A abolishes TGFbeta-induced antiproliferative and transcriptional responses, whereas depletion of PPM1A enhances TGFbeta signaling in mammalian cells. Smad-antagonizing activity of PPM1A is observed during Nodal-dependent early embryogenesis in zebrafish. PPM1A / PP2C-alpha plays a critical role in terminating TGFbeta signaling through dephosphorylation of Smad2/3. PPM1A / PP2C-alpha plays a critical role in the regulation of normal placentation by inhibiting trophoblast migration and invasion. PPM1A and to some extent PPM1B are important negative regulators of positive transcription elongation factor b (P-TEFb) function.
