PI3K/AKT pathway Background

PI3K/AKT pathway is a signaling pathway essential for apoptosis of cells. The signaling is activated by receptor tyrosine kinases(RTK) such as IGF1R. PI3K is a key member in this signaling pathway. It is classified into three types: typeⅠ, typeⅡ and type Ⅲ. Activated PI3K plays an important role in the process of PIP2 conversing into PIP3. Triple phosphoralation of PI is necessary for the binding of AKT/PKB to the cell membrane and its subsequent activation by PDK1. The key regulator is PTEN, a phosphatase that regulates PI3K and SHC phosphorylation. RAS may also regulate PI3K activity. AKT/PKB is a Mr approximately 60,000 serine/threonine kinase similary with PKA and PKC. AKT/PKB may directly phospholate various transcription factors to inhibit apoptotic gene expression. Through activated AKT, a number of key mitogenic processes are activated including inhibition of apoptosis, survival gene transcription, cell cycle progression, protein translation, and cell growth and proliferation. AKT/PBK can also Positive regulate two transcription factors: NF-κB and Bcl-2 and then regulate cell apoptosis. AKT can directly phosphorylate apoptotic protein Bad, which is an antagonist of Bcl-2 and Ecl-K, to promote cell apoptosis. In clinical, PI3K/AKT pathway is essential for proliferation and survival of melanoma cells.
