Using the leucine-rich repeats (LRRs) of the SLIT (see SLIT1; 603742) proteins in a database search, followed by RT-PCR of brain RNA, Lauren et al. (2003) cloned LRRTM4. The deduced 518-amino acid protein contains an N-terminal signal sequence, followed by 10 extracellular LRRs flanked by cysteine-rich domains, a transmembrane region, and an intracellular tail. LRRTM4 has 4 N-glycosylation sites, several putative phosphorylation sites, and a C terminus that ends in the sequence ECEV, which is a characteristic of LRRTM family members. Lauren et al. (2003) also identified mouse Lrrtm4, which shares 95% identity with human LRRTM4. RT-PCR analysis detected prominent LRRTM4 expression in human CNS, including cerebellum, amygdala, caudate nucleus, corpus callosum, hippocampus, thalamus, and spinal cord, and intermediate levels in human lung, muscle, prostate, salivary gland, small intestine, stomach, and testis. In developing mice, Lrrtm4 expression began at embryonic day 15, and highest levels were reached at postnatal day 1 and persisted into adulthood. In situ hybridization of adult mouse brain detected widespread Lrrtm4 expression predominantly in neurons with specific patterns for each Lrrtm.
