Junctional adhesion molecule C, also known as Junctional adhesion molecule 3, JAM3, is a single-pass type I membrane protein which belongs to the immunoglobulin superfamily. JAM3 is widely expressed. The highest expression of JAM3 is in placenta, brain and kidney. JAM3 contains one Ig-like C2-type (immunoglobulin-like) domain and one Ig-like V-type (immunoglobulin-like) domain. At the protein level JAM3 displays 36 and 32% identity to JAM2 and JAM1, respectively. JAM3 participates in cell-cell adhesion distinct from tight junctions. Tight junctions represent one mode of cell-to-cell adhesion in epithelial or endothelial cell sheets, forming continuous seals around cells and serving as a physical barrier to prevent solutes and water from passing freely through the paracellular space. JAM3 is localized in the tight junctions between high endothelial cells. Unlike other proteins in the same family, this protein is unable to adhere to leukocyte cell lines and only forms weak homotypic interactions.
