Influenza Neuraminidase (NA) Background
Neuraminidase is a single-pass type II membrane protein which exists as a homotetramer. It specifically catalyzes the hydrolysis removal of terminal sialic acid residues, and thus also called sialidase. Influenza neuraminidase (NA) promotes influenza virus release from infected cells and facilitates virus spread within the respiratory tract. At present, there are only three licensed anti-Influenza drugs namely Relenza (Zanamivir - ZMV), Tamiflu (Oseltamivir - OTV) and Amantadine/Rimantadine. The latter targets the M2 ion channel whereas the other compounds target neuraminidase and were designed through structure-based enzyme inhibitor programmes. The neuraminidase inhibitors interfere with the release of progeny influenza virus from infected host cells, thereby halts the spread of infection in the respiratory tract. The neuraminidase inhibitors are effective against all neuraminidase subtypes and, therefore, against all strains of influenza, a key point in epidemic and pandemic preparedness. In addition, The neuraminidase inhibitors are associated with very little toxicity and are far less likely to promote the development of drug-resistant influenza. Because of a broader antiviral spectrum, better tolerance, and less potential for emergence of resistance than is seen with the M2 inhibitors, the neuraminidase inhibitors represent an important advance in the treatment of influenza.
The viral neuraminidases are frequently used as an antigenic determinants found on the surface of the Influenza virus. Some variants of the influenza neuraminidase (NA) confer more virulence to the virus than others. Swiss-Prot lists 137 types of neuraminidase from various species as of October 18, 2006, while nine subtypes of influenza neuraminidase (NA) are known. Influenza neuraminidase (NA) exists as a mushroom-shape projection on the surface of the influenza virus. It has a head consisting of four co-planar and roughly spherical subunits, and a hydrophobic region that is embedded within the interior of the virus' membrane.
