Influenza A virus is capable of rapid genetic change in mammals. In humans, the need to escape preexisting immunity exerts positive selection pressure on changes in amino acids comprising the antigenic sites of the surface glycoproteins, hemagglutinin (HA) and neuraminidase (NA). The Hemagglutinin (HA) protein is synthesised in the infected cell as a single polypeptide chain, HA0. This initial molecule has to be cleaved by the host cell proteases into disulfide linked HA1 (47 kDa) and HA2 (29 kDa) subunits in order for the virus to mediate membrane fusion and subsequent infection. In October 1999, H4N6 influenza A viruses were isolated from pigs with pneumonia on a commercial swine farm in Canada. Phylogenetic analyses of the sequences of all eight viral RNA segments demonstrated that these are wholly avian influenza viruses of the North American lineage. This is the first report of interspecies transmission of an avian H4 influenza virus to domestic pigs under natural conditions. Other researchers found that Avian lineage H4N6 influenza viruses previously isolated from pigs differ at hemagglutinin amino acids 226 and 228 from H4 subtype viruses isolated from birds.
