IL7R / IL7RA Protein Description
Interleukin 7 Receptor alpha ( IL-7Rα ), also known as CD127, is a hematopoietin receptor superfamily member that plays an important role in lymphocyte differentiation, proliferation, and survival. IL-7Rα is majorly expressed on T cells and their precursors, and early in B cell development as well, prior to the appearance of surface IgM. Mature human IL-7Rα consists of a 219 aa extracellular domain (ECD) with one fibronectin type-III domain and a WSxWS motif, a 25 aa transmembrane segment, and a 195 aa cytoplasmic domain. Within the ECD, human IL-7Rα shares 67% aa sequence identity with mouse IL-7Rα. Mutations in the human IL-7Rα gene cause a type of severe combined immunodeficiency in which the major deficiencies are in T cell development, whereas B and NK cells are relatively normal in number. Therefore, the IL-7Rα is critically required for the proper development and function of lymphoid cells. In addition to IL-7, IL-7Rα also associates with TSLPR to form the functional receptor for thymic stromal lymphopoietin (TSLP) which indirectly regulates T cell development by modulating dendritic cell activation. Dynamic regulation of IL-7Rα is important for the generation of appropriate immune responses.
