Background
Human SEF (similar expression to fgf genes), also known as interleukin 17 receptor D (IL17 Rd), is a type I transmembrane receptor that shares sequence homology with the intracellular domain of members of the IL-17 receptor family. It is highly expressed in umbilical vein endothelial cells and in several highly vascularized tissues such as kidney, colon and heart. Two transcript variants resulting from alternative splicing exit, and the full-length long form is found in both the cytoplasm and plasma membrane. SEF is highly conserved in zebrafish, mouse, and humans, and studies in each system indicate that SEF acts as a feedback inhibitor of FGF-mediated Ras-MAPK signaling and ERK activation by inhibiting FGFR1 tyrosine phosphorylation and/or Ras downstream events. Additionally, SEF is also identified as a mediator of JNK and apoptosis by activating JNK through a TAK1-MKK4-JNK pathway. Human SEF is likely to play critical roles in endothelial or epithelial functions such as proliferation, migration, and angiogenesis.
