IL-4

Participates in at least several B-cell activation processes as well as of other cell types. It is a costimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes.

Summaries for IL-4

Wikipedia summary for IL-4:

Interleukin-4, abbreviated IL-4, is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. Upon activation by IL-4, Th2 cells subsequently produce additional IL-4. The cell that initially produces IL-4, thus inducing Th0 differentiation, has not been identified, but recent studies suggest that basophils may be the effector cell.[1] It is closely related and has functions similar to Interleukin 13. Its receptor is the Interleukin-4 receptor.

Sequence similarities:

Belongs to the IL-4/IL-13 family.

Subcellular location: Secreted.
Involvement in disease: Genetic variations in IL4 may be a cause of susceptibility to ischemic stroke (ISCHSTR) [MIM:601367]; also known as cerebrovascular accident or cerebral infarction. A stroke is an acute neurologic event leading to death of neural tissue of the brain and resulting in loss of motor, sensory and/or cognitive function. Ischemic strokes, resulting from vascular occlusion, is considered to be a highly complex disease consisting of a group of heterogeneous disorders with multiple genetic and environmental risk factors.

  • IL-4 is a important switch factor directing Ig-E synthesis
  • IL-4 T cell and mast cell growth factor activities
  • IL-4 potent modulator of ion transport in the bronchial epithelium and of ion composition of airway surface fluid
  • inhibition of matrix metalloproteinas-3 (MMP-3) synthetis in fibroblasts
  • IL-4 is involved in excessive deposition of extracellular matrix (EMC) proteins in giant papillae
  • IL-4 directing germinal center-B cells to differentiate into memory B cells (Zhang 2001)
  • Th2-promoting cytokine IL4 played a permissive role that superseded Treg function, thereby allowing the development of allergic inflammation
  • IL-4 with its STAT6-signalling pathway are critical for the expression of Th2 effector immune responses in peripheral tissues such as the skin, lung and gut
