| Western blot : | This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Mouse IFNAR1 in WB. Using a DAB detection system, the detection limit for Mouse IFNAR1 is approximately 0.25 ng/lane under non-reducing conditions and reducing conditions. |

IFNAR-1 Antibody Background
Interferon alpha/beta receptor 1, also known as IFN-alpha/beta receptor 1, Type I interferon receptor 1, Cytokine receptor family 2 member 1, Cytokine receptor class-II member 1, CRF2-1, IFNAR1 and IFNAR, is a single-pass type I membrane protein which belongs to thetype II cytokine receptor family. IFNAR1 is a receptor for interferons alpha and beta which contains threefibronectin type-III domains. The binding of IFNAR1 to type I IFNs triggers tyrosine phosphorylation of a number of proteins including JAKs, TYK2, STAT proteins and IFNR alpha- and beta-subunits themselves. IFN receptors are present in all tissues and even on the surface of most IFN-resistant cells. Isoform1, isoform2and isoform3are expressed in the IFN-alpha sensitive myeloma cell line U266S. Isoform2and isoform3are expressed in the IFN-alpha resistant myeloma cell line U266R. Ubiquitination, endocytosis, and lysosomal degradation of the IFNAR1 subunit of the type I interferon (IFN) receptor is mediated by the SCFbeta-Trcp (Skp1-Cullin1-F-box protein beta transducin repeat-containing protein) E3 ubiquitin ligase in a phosphorylation-dependent manner. Stability of IFNAR1 is regulated by its binding to Tyk2 kinase. Expression of Tyk2 stabilized IFNAR1 in a manner that was dependent neither on its binding to beta-Trcp nor IFNAR1 ubiquitination.
