Human IL12B Materials provided
Capture Ab:1 mg/mL of rabbit anti-Human IL12B / IL-12B monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 1 μg/mL in PBS before coating. (Catalog: # 10052-MM05)
Detection Ab:0.2 mg/mL of mouse anti-Human IL12B / IL-12B monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4, store at 4℃). Dilute to working concentration of 0.4 μg/mL in detection antibody dilution buffer before use. (Catalog: # 10052-R054)
Standard:Each vial contains 24 ng of recombinant Human IL12B / IL-12B. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 1000 pg/mL is recommended.
Human IL12B Sensitivity
The minimum detectable dose of Human IL12B / IL-12B was determined to be approximately 15.625 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.
Human IL12B Principle of the product
The Human IL12B / IL-12B ELISA Pair Set is for the quantitative determination of Human IL12B / IL-12B.
The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for Human IL12B / IL-12B coated on a 96-well plate. Standards and samples are added to the wells, and any Human IL12B / IL-12B present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated mouse anti-Human IL12B / IL-12B monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of Human IL12B / IL-12B present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.
Human IL12B Storage

IL12B/IL-12B Background

Subunit beta of interleukin 12 (also known as natural killer cell stimulatory factor 2, or cytotoxic lymphocyte maturation factor 2, p40) (IL12B) is a subunit of human interleukin 12. IL12B/IL-12B is a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. Interleukin 12 is a disulfide-linked heterodimer composed of the 40 kD cytokine receptor like subunit encoded by this gene, and a 35 kD subunit encoded by IL12A. IL12B/IL-12B is expressed by activated macrophages that serve as an essential inducer of Th1 cells development. This cytokine has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen. Overexpression of this gene was observed in the central nervous system of patients with multiple sclerosis (MS), suggesting a role of this cytokine in the pathogenesis of the disease. The promoter polymorphism of this gene has been reported to be associated with the severity of atopic and non-atopic asthma in children. IL12B/IL-12B associates with IL23A to form the IL-23 interleukin, an heterodimeric cytokine which functions in innate and adaptive immunity.
