The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography. |

HAI-1 / SPINT1 Antibody Background
Kunitz-type protease inhibitor 1, also known as hepatocyte growth factor activator inhibitor type 1, HAI-1 and SPINT1, is a secreted protein which belongs to the Kunitz family of serine protease inhibitors. SPINT1 / HAI-1 contains twoBPTI / Kunitz inhibitor domains, oneLDL-receptor class A domain and oneMANSC domain. SPINT1 / HAI-1 is an epithelial Kunitz-type transmembrane serine protease inhibitor that is encoded by the SPINT1 gene. SPINT1 / HAI-1 is a potent inhibitor specific for HGF activator and is thought to be involved in the regulation of the proteolytic activation of HGF in injured tissues. Alternative splicing results in multiple variants encoding different isoforms. SPINT1 / HAI-1 is a membrane-bound, serine proteinase inhibitor initially identified as an inhibitor of hepatocyte growth factor activator. It displays potent inhibitory activity toward a large number of trypsin-like serine proteases. SPINT1 / HAI-1 is shown to play an essential role in postnatal epithelial homeostasis. SPINT1 / HAI-1 has critical roles in the regulated keratinization of the epidermis and hair development. It inhibits matriptase and prostasin, both of which are membrane-bound serine proteinases that have critical roles in epidermal differentiation and function. Matriptase suppression is an essential function of SPINT1 / HAI-1 in postnatal tissue homeostasis.
