Preparation :

This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human HAI-1 / SPINT1 (rh HAI-1 / SPINT1; Catalog#11742-H08H; O43278-2; Met 1-Val 433). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

Direct ELISA : This antibody can be used at 0.5-1 μg/mL with the appropriate secondary reagents to detect Human SPINT1. The detection limit for Human SPINT1 is approximately 0.0156 ng/well.
Storage : This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

HAI-1 / SPINT1 Antibody Background

Kunitz-type protease inhibitor 1, also known as hepatocyte growth factor activator inhibitor type 1, HAI-1 and SPINT1, is a secreted protein which belongs to the Kunitz family of serine protease inhibitors. SPINT1 / HAI-1 contains twoBPTI / Kunitz inhibitor domains, oneLDL-receptor class A domain and oneMANSC domain. SPINT1 / HAI-1 is an epithelial Kunitz-type transmembrane serine protease inhibitor that is encoded by the SPINT1 gene. SPINT1 / HAI-1 is a potent inhibitor specific for HGF activator and is thought to be involved in the regulation of the proteolytic activation of HGF in injured tissues. Alternative splicing results in multiple variants encoding different isoforms. SPINT1 / HAI-1 is a membrane-bound, serine proteinase inhibitor initially identified as an inhibitor of hepatocyte growth factor activator. It displays potent inhibitory activity toward a large number of trypsin-like serine proteases. SPINT1 / HAI-1 is shown to play an essential role in postnatal epithelial homeostasis. SPINT1 / HAI-1 has critical roles in the regulated keratinization of the epidermis and hair development. It inhibits matriptase and prostasin, both of which are membrane-bound serine proteinases that have critical roles in epidermal differentiation and function. Matriptase suppression is an essential function of SPINT1 / HAI-1 in postnatal tissue homeostasis.
