FLRG Antibody Background
FLRG (follistatin-related gene) protein is a follistatin-like protein which contains two FS domains and acts as a binding protein for activin and BMP-2. FLRG is a secreted glycoprotein that is highly homologous with follistatin. These proteins are involved in the regulation of various biological effects mediated by their binding to TGF-β superfamily members, activin A and bone morphogenetic proteins.The mouse FLRG gene consists of five exons, and each encodes discrete functional regions.Although mouse and human FLRGs show some divergence compared with follistatins, structurally they are conserved, also, domain organization and putative Asn-linked glycosylation sites are completely conserved. Both mouse and human FLRG have an NH2-terminal domain, two FS domains that are thought to have growth factor-binding motifs, and a COOH-terminal acidic domain. Recombinant mouse FLRG protein binds to activin A and BMP-2, and blocked the activities of acitvin A, activin B, activin AB, and BMP-2. Mouse FLRG mRNA is abundantly expressed in heart, lung, kidney, and testis. FLRG is also expressed in the mouse ovary, suggesting that FLRG plays physiological roles in the regulation of ovarian function as follistatin does.
