Fibroblast growth factor receptor 1 (FGFR1) is a member of the fibroblast growth factor receptor (FGFR) family, whose amino acid sequence is highly conserved between members throughout evolution. FGFR family members differ from one another in their ligand affinities , tissue distribution and have been implicated in a multitude of activities. FGFR1, also known as fms-related tyrosine kinase-2 (FLT2) and CD331, is a receptor tyrosine kinase whose ligands are specific members of the fibroblast growth factor family. This protein structurally consists of an extracellular region containing three immunoglobulin-like domains, a single hydrophobic membrane-spanning segment and a cytoplasmic tyrosine kinase domain. The extracellular portion of the protein interacts with fibroblast growth factors, setting in motion a cascade of downstream signals, ultimately influencing mitogenesis and differentiation. FGFR1 is the receptor for FGF basic and also interacts with GRB14, SHB, FRS2. Defects in FGFR1 are the cause of several deseases, such as Kallmann syndrome type 2 (KAL2), osteoglophonic dysplasia (OGD), as well as Pfeiffer syndrome (PF).
