EphB2 / EPHB2 Antibody Background
EphB2, also known as Cek5, Nuk, Erk, Qek5, Tyro5, Sek3, Hek5, and Drt, is a member of the transmembrane Eph receptor tyrosine kinase family (RTKs). Ephrins are divided into the ephrin-A class, anchored to the membrane by a glycosylphosphatidylinositol linkage, and the ephrin-B class, the transmembrane proteins. The Eph receptor family are divided into two groups based on the similarity of their extracellular domain sequences and affinities for binding ephrin-A and ephrin-B ligands, making up the largest subgroup of the receptor tyrosine kinase (RTK) family. Human EphB2 cDNA encodes a 1055 aa precursor, which includes an 18 aa signal sequence, a 525 aa extracellar domain (ECD) containing a cysteine rich region followed by two fibronectin type I II domains, a 21 aa transmembrane segment, and a 491 aa cytoplamic domain containing the tyrosine kinase domain, a sterile alpha motif (SAM), and a PDZ binding motif. It is expressed on both sides of the neuronal synapse controlling axon guidance across the embryonic midline, promoting a neuronal fate from neural precursors, and regulating NMDA receptor activity. EphB2 is a receptor for members of the ephrin-B family, and is proposed as a tumor suppressor. It is indicated that aberrant EphB2 expression and activity are implicated in the progression of several cancers.
