Ebola virus Matrix protein VP24 / Ebola virus VP24 Background

VP24 is a minor matrix protein of Ebola virus, which plays an important role in nucleocapsid assembly, virus release and replication. It is resistant to trypsin digestion in the absence of Triton X-100. When expressed in mammalian cells, it prefers to oligomerize into tetramers. The oligomerization ability is believed to be critical for the role of VP24 in nucleocapsid assembly; it is considered that the N-terminal region of VP24, especially the amino acids 90 to 130, is responsible for oligomer formation. VP24 is identified present in virus-like particles produced by VP24-expressing mammalian cells. Recent studies demonstrate that VP24 also acts as a suppressor of immune system during the early stages of host defense. VP24 can bind to karyopherins such as karyopherin α1, which transports the transcription factor STAT1 to the nucleus, inhibiting signaling downstream of interferon (IFN). Moreover, VP24 can block the p38-α stimulated by IFNβ, preventing the signal transduction in MKK3/6-p38 mitogen-activated protein (MAP) kinase pathway. In addition, direct binding between VP24 and STAT1 is discovered, which may result in STAT1 degradation or the failure of STAT1 phosphorylation, and thus suppress the host immune response. There will be attempts to design novel anti-Ebola virus therapeutics by impairing the multiple functions of VP24.
