Background
Dipeptidyl-peptidase II (DPPII), also known as quiescent cell proline dipeptidase (QPP) and dipeptidyl-peptidase 7 (DPP7), is a post-proline cleaving amino peptidase expressed in quiescent lymphocytes. As an intracellular protease localized to the vesicular system, it releases preferably N-terminal dipeptides from oligopeptides with Pro or Ala in the penultimate position at acidic pH.. The proline-specific DPPs like DPP7, fibroblast activation protein alpha (FAP), DPPIV, DPP8 and DPP9, on the basis of their preference for cleavage after X-Pro, are likely to be involved in Pro-specific N-terminal processing regulating an increasing number of biological processes. These DPPs are emerging as an important protease family involved in the regulation of signaling by peptide hormones. The purified rhDPP7 is active against Lys-Pro-AMC and Lys-Ala-AMC. Its activity against Lys-Pro-AMC is approximately 10-fold of that against Lys-Ala-AMC under identical conditions.
