NADH-cytochrome b5 reductase 1 (CYB5R1) belongs to the flavoprotein pyridine nucleotide cytochrome reductase family. This reductase contains 1 FAD-binding FR-type domain and is involved in desaturation and elongation of fatty acids, cholesterol biosynthesis, drug metabolism, and, in erythrocyte, methemoglobin reduction. Cytochrome b5 reductase, a flavoprotein that has an NADH binding site, can provide electrons to p450 monooxygenases through cytochrome b5. Reactions catalyzed by P450 show that the properties of the associated electron transport proteins that support P450-1 activity in mutants are consistent with cytochroe b5: NADH-cytochrome b5 oxidoreductase (cyt b5: cyt b5 reductase). Experiments have indicated that a flavoprotein is involved in the reaction that GA12 synthesis from GA12-aldehyde and consistent with participation of cytochrome b5 reductase which has an FAD prosthetic group. Added cytochrome b5 would enhance coupling of cytochrome b5 reductase to the P450-1 monooxygenase facilitating electron transport, while added FAD would restore bound flavin that may have been lost from the cyt b5 reductase during microsome preparation.
