CSNK1G2 Antibody Background
Casein kinase I gamma 2 isoform (CSNK1G2), a member of the large casein kinase I (CKI) family, may affect the development of brain, and associate with vesicular trafficking and neurotransmitter releasing from small synaptic vesicles. Casein kinases are operationally defined by their preferential utilization of acidic proteins such as caseins as substrates. It can phosphorylate a large number of proteins, and participates in Wnt signaling. The Casein kinase 1 family of protein kinases are serine/threonine-selective enzymes that function as regulators of signal transduction pathways in most eukaryotic cell types. The CKI family includes several other isoforms (alpha, beta, gamma, and delta). CKIgamma2 (CSNK1G2) hyperphosphorylates the serine-repeat motif of CERT, thereby inactivating CERT and down-regulating the synthesis of sphingomyelin. MTA1s interacts with CSNK1G2 both in vitro and in vivo and colocalizes in the cytoplasm. MTA1s as a target of CSNK1G2, and provided new evidence to suggest that CSNK1G2 phosphorylates and modulates the functions of MTA1s. In mammalian cells, CSNK1G2 selectively and constitutively binds Smad3 but not Smad1, -2 or -4. CSNK1G2 is the first kinase known to mark activated Smad3 for destruction. Because the overexpression in human cancers, CSNK1G2 may act as an oncoprotein during tumorigenesis.
