CSF1R / M-CSFR / CD115 Protein Description

Macrophage colony-stimulating factor 1 receptor, also known as M-CSFR, CSF-1-R, and CD115, is a single-pass type I  membrane protein. M-CSFR encoded by the proto-oncogene c-fms is the receptor for colony stimulating factor 1, a cytokine involved in the proliferation, differentiation, and activation of macrophages. As a member of the type I ¢ subfamily of receptor tyrosine kinases (RTKs), M-CSF receptor / CD115 is expressed mainly on cells of monocyte/macrophage lineage, and consists of an extracellular ligand-binding domain and a cytoplasmic protein tyrosine kinase (PTK) domain joined through a single transmembrane helix. Ligand binding induces the M-CSF receptor homodimerization, and leads to transphosphorylation of specific tyrosine and the subsequent signal transduction. This activated receptor exerts actions on multiple intracellular effectors which determine the magnitude and specificity of the biological responses, and thus mediates most if not all of the biological effects of the CSF1 cytokine, including cell growth, survival, differentiation and chemotaxis. Defects in M-CSFR are associated with chronic myelomonocytic leukemia and type M4 acute myeloblastic leukemia. Increased levels of M-CSFR are found in microglia in Alzheimer's disease and after brain injuries. The increased receptor expression causes microglia to become more active.
