CD95 / APO-1 / TNFRSF6 / FAS Antibody Background

Fas, also known as APO-1, CD95 and TNFRSF6, is a member of the tumor necrosis factor (TNF) / nerve growth factor (NGF) receptor superfamily. Fas is highly expressed on the surface of a number of cell types including epithelial cells, hepatocytes, activated mature lymphocytes and other tumor cells. This receptor contains a death domain and has been shown to play a central role in the physiological regulation of programmed cell death (PCD, apoptosis) upon ligation of Fas ligand (Fas-L). This Fas receptor has been reported to be involved in the activation of NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and the alternate pathways for CTL-mediated cytotoxicity. Accordingly, this protein is implicated in the pathogenesis of various malignancies and diseases of the immune system. Multiple Fas isoforms are derived from alternative spliced transcript variants, and among which, a soluble form lacking the transmembrane domain may negatively regulate the apoptosis mediated by the full length isoforms.
