Background

As a transmembrane protein, CD7 is a member of the immunoglobulin superfamily. It is normally expressed on all T-lymphocytes, NK-cells, pre-B lymphocytes and pleuripotent hematopoietic stem cells. N-terminal amino acids 1-107 of CD7 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain of CD7 is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 plays an essential role in T-cell interactions, T-cell/B-cell interaction during early lymphoid development, T- and NK-cell activation and cytokine production. The regulation of CD7 gene expression through NF-kappaB activation induced by TCR/CD28 might have significant implications for T-cell homeostasis. CD7 has been shown to interact with PIK3R1. The type I I PI 4-kinases are integral components of the CD7 signaling pathway. Recently, an epithelial cell secreted protein, K12, was identified as a CD7 ligand. Gal-3 might bind to CD7 to induce apoptosis. Gal-1, another galectin sharing high homology with Gal-3, may also cause apoptosis in T cells through binding to CD7. The CD7 expression was an independent risk factor for disease free (DFS) and post-remission survivals (PRS), and it is also involved in chronic myeloid leukemia (CML) and adult T-cell lymphoma/leukaemia (ATLL). Nevertheless, the function of the CD7 protein in the immune system is largely unknown.
