Preparation:This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human CD40 / TNFRSF5 (rh CD40 / TNFRSF5; Catalog#10774-H08H; NP_001241.1; Met1-Arg193). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

FCM: 0.5-2 μg/Test

ICC/IF: 10-25 μg/mL

Storage:This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -80℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

Caption:
Immunofluorescence staining of Human CD40 in Hep G2 cells. Cells were fixed with 4% PFA, blocked with 10% serum, and incubated with Mouse anti-Human CD40 monoclonal antibody (15 µg/ml) at 37℃ 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-mouse IgG secondary antibody (left panel, captured by laser confocal scanning microscope; right panel, captured by fluorescence microscope), countstained with DAPI (blue). Positive staining was localized to cytoplasm and cytomembrane.

Human whole blood lymphocytes were stained with purified anti-Human CD40, then a FITC-conjugated second step antibody. The histogram were derived from gated events with the forward and side light-scatter characteristics of viable lymphocytes.

                CD40/TNFRSF5 Background

                CD40, also known as TNFRSF5, is a member of the TNF receptor superfamily which are single transmembrane-spanning glycoproteins. CD40 protein plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. CD40 protein is expressed in B cells, dendritic cells, macrophages, endothelial cells, and several tumor cell lines. Defects in CD40 result in hyper-IgM immunodeficiency type 3 (HIGM3). In addition, CD40/CD40L interaction is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis.
