Background
CD34 molecule is a member of a family of single-pass transmembrane sialomucin proteins, and is selectively expressed on hematopoietic progenitor cells and the small vessel endothelium of a variety of tissues. As a monomeric cell surface antigen, CD34 is highly glycosylated with nine potential N-linked and numerous potential O-linked glycosylation sites in its extracellular domain. Although used clinically in hematopoietic stem cell transplantation for more than 15 years, little is known about the exact function of CD34. It may act as an adhesion molecule in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells, and is also required for T cells to enter lymph nodes via interaction with L-selectin. It was indicated that CD34 is a phosphorylation target for activated PKC, and couples to the hematopoietic adapter protein CrkL, which were involved in CD34 signaling pathways. CD34 is abberantly expressed in many kinds of tumors and is implicated in leukemogenesis.
