Macrophage Marker Background
Macrophages are produced by the differentiation of monocytes. Monocytes leave the bone marrow, circulate through the blood steam, and enter the tissues through a mechanism called leukocyte extravasation where they mature into macrophages. Both macrophages and monocytes are phagocytes, acting in both non-specific defense (innate immunity) as well as to help initiate specific defense mechanisms (adaptive immunity) of vertebrate animals. Macrophages and monocytes phagocytose (engulf and then digest) cellular debris and pathogens either as stationary or as mobile cells, and to stimulate lymphocytes and other immune cells to respond to the pathogen.
Macrophages can be identified by specific expression of a number of proteins including CD14, CD11b, F4/80 (mice)/EMR1 (human), CD68 and MAC-1/MAC-3 by flow cytometry or immunohistochemical staining. One of the functionally important receptors in macrophage biology is PECAM-1 (CD31), which, through homophilic adhesion, determines which cells will be phagocytosed.
