CAMKI (CAMK1)

CaM-kinase II (CAMK2) is a prominent kinase in the central nervous system that may function in long-term potentiation and neurotransmitter release. Member of the NMDAR signaling complex in excitatory synapses it may regulate NMDAR-dependent potentiation of the AMPAR and synaptic plasticity

Summaries for CAMKI/CAMK1

Entrez Gene summary for CAMKI/CAMK1:

Calcium/calmodulin-dependent protein kinase I is expressed in many tissues and is a component of a calmodulin-dependent protein kinase cascade. Calcium/calmodulin directly activates calcium/calmodulin-dependent protein kinase I by binding to the enzyme and indirectly promotes the phosphorylation and synergistic activation of the enzyme by calcium/calmodulin-dependent protein kinase I kinase.

OMIM - description for CAMKI/CAMK1:

Calcium/calmodulin (see CALM1; 114180)-dependent protein kinases (CAMKs) are a subfamily of the serine/threonine protein kinase family. Multiple substrates for rodent Camk1 have been identified, including SYN1 (313440), SYN2 (600755), CREB (see 123810), and CFTR (602421). Camk1 activity is dependent upon its phosphorylation by a Camk1 kinase. By screening an HL-60 cDNA library using a neutrophil-derived protein kinase-like clone as the probe, Haribabu et al. (1995) isolated a cDNA encoding human CAMK1. The predicted 370-amino acid human CAMK1 protein shares 97% amino acid identity with rat Camk1. Northern blot analysis detected a widely expressed CAMK1 transcript of 1.5 kb.

Wikipedia summary for CAMKI/CAMK1:

Calcium/calmodulin-dependent protein kinase type 1 is an enzyme that in humans is encoded by the CAMK1 gene.Calcium/calmodulin-dependent protein kinase I is expressed in many tissues and is a component of a calmodulin-dependent protein kinase cascade. Calcium/calmodulin directly activates calcium/calmodulin-dependent protein kinase I by binding to the enzyme and indirectly promotes the phosphorylation and synergistic activation of the enzyme by calcium/calmodulin-dependent protein kinase I kinase

Human CAMKI/CAMK1 Protein General Information

Protein names

Recommended name: Calcium/calmodulin-dependent protein kinase type II subunit alpha Short name=CaM kinase II subunit alpha Short name=CaMK-II subunit alpha

Sequence length

478 AA.

Enzyme regulation

Autophosphorylation of Thr-286 allows the kinase to switch from a calmodulin-dependent to a calmodulin-independent state

Sequence similarities:

Belongs to the protein kinase superfamily. CAMK Ser/Thr protein kinase family. CaMK subfamily. Contains 1 protein kinase domain.

Subunit structure

CAMK2 is composed of four different chains: alpha, beta, gamma, and delta. The different isoforms assemble into homo- or heteromultimeric holoenzymes composed of 8 to 12 subunits. Interacts with BAALC, MPDZ, SYN1, CAMK2N2 and SYNGAP1

Subcellular location: Cell junction › synapse › presynaptic cell membrane By similarity. Cell junction › synapse By similarity. Note: Postsynaptic lipid rafts

Genatlas:

  • CAMKI/CAMK1 is involved in a number of cellular processes like transcriptional regulation, hormone production, translational regulation, regulation of actin filament organization and neurite outgrowth
  • CAMKI/CAMK1 is involved in calcium- dependent activation of the ERK pathway
  • CAMKI/CAMK1 is potentially implicated in the regulation of bone and cartilage development
  • CAMKI/CAMK1 can regulates the functions of RPS19 through phosphorylation of Ser59
  • CAMKI/CAMK1 recruite Ca(2+)-permeable AMPARs which may provide a mechanistic link between NMDAR activation in long-term potentiation and regulation of a signaling pathway that drives spine enlargement via actin polymerization
