BMP and activin membrane-bound inhibitor homolog, also known as BAMBI and NMA, is a single-pass type I  membrane protein which belongs to the BAMBI family. Members of the transforming growth factor-beta (TGF-beta) superfamily, including TGF-beta, bone morphogenetic proteins (BMPs), activins and nodals, are vital for regulating growth and differentiation. These growth factors transduce their signals through pairs of transmembrane type I  and type I I receptor kinases. BAMBI is a transmembrane glycoprotein related to the transforming growth factor-beta (TGF-beta)-family type I  receptors and functions as a negative regulator of TGF-beta signaling during development. BAMBI is co-expressed with the ventralizing morphogen BMP4 during Xenopus embryogenesis and that it requires BMP signalling for its expression. BAMBI is induced by BMP signaling through the evolutionary conserved BMP-responsive elements in its promoter. Wnt/beta-catenin signaling activates transcription of BAMBI and that BAMBI expression is aberrantly elevated in most colorectal carcinomas. BAMBI stably associates with TGF-beta-family receptors and inhibits BMP and activin as well as TGF-beta signalling. BAMBI negatively regulates TGF-beta-family signalling by a regulatory mechanism involving the interaction of signalling receptors with a pseudoreceptor.
