The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography. |

HAVCR1 / TIM1 / KIM1 Antibody Background
Human hepatitis A virus (HAV) is a hepatotropic virus that causes acute hepatitis in humans, and its target receptor HAVCR1 is encoded by an important asthma and allergy susceptibility gene in humans. The monkey and human HAVCR1 are the first identified members of the T-cell immunoglobulin mucin (TIM) family, an immunologically important group of receptors that is conserved in vertebrates. Human HAV cellular receptor 1 (HAVCR1), also known as KIM1 and TIM1, is widely expressed with highest levels in kidney and testis. It has been shown that HAVCR1 is expressed on activated CD4+T-cells during the development of helper T-cells responses and associates with immunomodulatory properties. TIM1 is a type 1 integral membrane glycoprotein, and plays a critical role in regulating T-cell differentiation and the development of atopy. It consists of a characteristic six-cysteine Ig-like domain (D1) extended above the cell surface by a mucin-like domain that contains a variable number of threonine, serine, and proline (TSP) hexameric repeats. D1 and its first N-glycosylation site are required for binding of HAV to HAVCR1. It also exists as a soluble form constitutive cleavage by an undefined MMP. In addition to HAV, another identified ligand for HAVCR1 is TIMD4.
